Adjuvant Combination for Kidney Cancer Gets FDA's Blessing

The US Food and Drug Administration (FDA) has granted approval to the combination of belzutifan and pembrolizumab as an adjuvant treatment for patients with high-risk renal cell carcinoma (RCC) following nephrectomy. This decision is based on the results of the phase 3 CLEAR-3 trial, which demonstrated that the combination of belzutifan, an inhibitor of the hypoxia-inducible factor (HIF) pathway, and pembrolizumab, a programmed death-1 (PD-1) inhibitor, significantly reduced the risk of disease recurrence or death compared to placebo. The trial enrolled 431 patients with high-risk RCC who had undergone nephrectomy, and the primary endpoint was met with a 47% reduction in the risk of disease recurrence or death in the belzutifan and pembrolizumab arm compared to the placebo arm. This approval represents a major advancement in the treatment of kidney cancer, offering patients a new therapeutic option that has the potential to improve survival rates and quality of life.
